Horse chestnut (Aesculus hipocastanum L.) belongs to horticulturally valuable woody species. Successfulness of its planting in Europe has decreased due to a pest invasion, the horse chestnut leaf miner Cameraria ohridella Deschka & Dimic, 1986. Research objective of this work was to induce organogenesis in explants taken from seedlings of horse chestnut inividuals. Explants (shoot tips of vegetative buds, petiole and shoot segments) collected in the winter and the summer were cultured on WPM and MS medium. The endogenous bacterial and yeast contamination was showed to be limiting factor of regeneration induction in the winter buds. In spite of application of wide-spectrum antibiotics (Hygromycin, Timentin, Cefotaxim) into the medium and bud surface treatment using Antibiotic-Antimycotic Stabilized suspension and Amphotericin B, no viable in vitro culture was derived. However shoot tips of summer buds were found as the most suitable and responsive explants with none or low rate of microbial inection (13%) and high proliferation rate (up to 80%). Benzyladenine (BA) and naphtylacetic acid (NAA) were positive factors of organogenesis induction.
