In this study, we describe the use of DNA analysis by RAPD method (Random Amplified Polymorphic DNA) to identify a variability of Norway spruce hurst ecotype. Total genomic DNA was isolated by DNA Plant Mini Kit (QIAGEN) from derived embryogenic cell lines. Embryogenic cultures were established from valuable original of hurst ecotype of Norway spruce (Picea abies [L.] Karst.) growing in the České Švýcarsko National Park and the Křivoklátsko Protected Landscape Area. From this area 39 samples of Norway spruce hurst ecotype were analysed, and 3 samples of Norway spruce mountain ecotype from locality Neveklov were compared. Arbitrary decamer primers from sets OPR, OPC and OPBC were tested. Polymorphism of observed Norway spruce lines was proved with primer OPBC – 17. The total of 420 scoreable fragments was amplified. 86 of them were monomorphic and 334 were polymorphic (79.5% polymorphism). The dendrogram of genetic similarity was constructed by the UPGMA method based on the Jaccard coefficient.
