The Simple Sequence Repeats (SSR) method of DNA analyses were used to determine the polymorphism of Norway spruce (Picea abies /L./ Karsten) samples. Total genomic DNA was extracted by DNA Plant Mini Kit (QIAGEN) from embryogenic cell lines of valuable original hurst ecotype of Norway spruce growing in the České Švýcarsko National Park and the Křivoklát Protected Landscape Area, and from needles taken from mountain ecotype of Norway spruce growing in Fláje (Ore Mountains) and Trčkov – Šerlišský kotel (Eagle Mountains), Czech Republic. Samples were screened using selected six polymorphic nuclear microsatellite markers. Measuring the size of amplification products was carried out using the genetic analyzer Applied Biosystems 3500. The obtained data were analyzed using the statistical program GenAlEx 6.5 (PEAKALL, SMOUSE 2006, 2012). The identified genetic loci were verified as highly polymorphic and could be further used for comparative genetic analyses of Norway spruce populations.
