Dormant axillary buds of endangered Betula nana were used for tissue culture establishment. Optimal cultivation media for organogenesis induction, multiplication and rooting were established. The modified agar medium WPM (LLOYD, MCCOWN 1980), with concentrations of phytohormones BAP 0.2 mg l-1 and IBA 0.1 mg l-1 was used for induction of birch organogenesis. The effect of different cytokinins and agars on multiplication was tested. The best results we obtained on WPM medium with concentrations of phytohormones BAP 0.2 mg l-1 and IBA 0.1 mg l-1, 200 mg l-1 of glutamine, 200 mg l-1 of casein hydrolysate, 30 g l-1 of saccharose, and 6 g l-1 of agar from company ROTH, pH adjusted to 5.8. Propagated shoots rooted in the agar WPM medium with concentration IBA (0.5 mg l-1). 100% of rooting rate of microcuttings was reached in this condition. The mortality during acclimatization was not over 2%.
