Protocol for in vitro propagation of critically endangered species Jurinea cyanoides (L.) Rchb. in the Czech Republic was established. The explant cultures were derived from vegetative shoots and seeds of endangered donor plants growing in open vegetation on the sandy dune in the isolated locality near Tišice in the Central Bohemia. The present number of these species individuals is so low that their natural reproduction is not guaranteed. In vitro cultivation is recognized as one of the important tools in the ex situ conservation. The induction of organogenesis and multiplication was successful on the modified MS medium (MURASHIGE, SKOOG 1962) with the concentration of substances 0.2 or 0.5 mg.l-1 of BAP and 0.1 mg.l-1 of IBA, 200 mg.l-1 of glutamine, 200 mg.l-1 of casein hydrolysate, 2 mg.l-1 of glycine, 30 g.l-1 of sucrose, 6 g.l-1 of agar, pH adjusted to 5.8. Rooting of microcuttings was reached on the 1/3 concentration of basal MS with enhanced concentration of auxin 3 mg.l-1 of IBA. The aim of this study was to initiate in vitro cultures to obtain large numbers of regenerated plantlets without disturbing the natural population. The preservation program was running in the cooperation with the Agency of Nature Protection.
